Reconstruction of Brankovice Parish

This is a reconstruction of the parish in Brankovice. The building has two floors, is partly a basement, attic, unused, false hip roof. The building was built in the Baroque - Classical style around 1773. The facade is horizontally divided by plinth, shams around windows, and by a cordon cornice lisenas vertically between windows. The front is accentuated by protruding buttress. The building consists of mid-section and two side wings, the layout is strictly symmetrical. The central axis of the main entrance to the building, which is linked to the entrance hall. On the second floor, there is a staircase hall in the central part of the building, connected to all rooms, which are further connected to each other. The rooms on the first floor will be used as an apartment for the pastor. The part of solution is reconstruction and extensions of bathrooms and garage. Bearing system of solved object consists of burnt bricks. The extensions is designed as ceramic brick blocks Therm masonry.
Implementation of Roof Construction at Koliště 47

The subject of the bachelor thesis is the realization of the roofing of the building Koliště 47. This building is located in the centre of Brno. The thesis deals with the solution of the truss system with a strip dormer and partly truss construction. Furthermore, the roofing made of folded aluminium sheet. The contents include a technical report, broader relations of transport routes, technological regulations, the solution of the site equipment, the design of the machine assembly, inspection and test plan, work safety for the specified technological stage and itemized budget.
Synchronous scene recording with multiple cameras

This thesis provides a detailed description of the basic equipment for synchronous capture from two cameras. The cameras used are thoroughly characterized, and the principles of their usage in scene capturing are explained. Furthermore, the theory of image fusion is examined, with a specific focus on fusing color and grayscale images from a thermal camera. The main emphasis is placed on the software framework, which is extensively described from camera initialization and image acquisition processes to storing captured images, fusion calibration, and the actual creation of the resulting fused image. The architecture of the application and its surface logic are outlined, along with a discussion of the key libraries utilized in the program and their significance for the entire process. This work establishes a stable software foundation for developing a program that enables synchronous capture and fusion of resulting images, thereby opening up new possibilities for image processing and analysis
Acute effects and addictive potential of new synthetic drugs from cathinone group - an animal study

Novel psychoactive substances (NPS) are a novel problem of the drug scene. NPS mimic effects of the "classic" illicit drugs, but since they have a different chemical structure, they are usually not covered by legislative control. The exact nature of the aforementioned effects depends merely on the description by users, with a proper scientific assessment still absent. Aim of this study is to evaluate effects and addictive potential of naphyrone (a derivative of cathinone) in Wistar rats. High concentration of naphyrone in the brain tissue discovered by pharmacokinetic analysis proved its high blood-brain barrier permeability. Brain level of naphyrone peaked at approximately 30 min after the treatment, nearly at the same time as in the serum. Since naphyrone administration significantly rises body temperature and increases overall locomotion, its stimulant effect is prominently apparent. Our study failed to prove any effect of naphyrone on sensorimotor gating. Tendency to produce conditioned place preference was observed but was not significant. My thesis reports on initial and novel findings about impact of naphyrone administration on physiological parameters of the animal model.
Effect of polyhistidine modification of viral particles on their intracellular localization and gene delivery to the nucleus

Viral vectors derived from mouse polyomavirus are a convenient tool for studying the targeted delivery of therapeutical agents into the cells and cellular organelles. Vectors derived from mouse polyomavirus face difficulties similar to other nanoparticles, as they often end up trapped inside an endosome where they are subsequently degraded. This diploma explored the potential of vector modifications, which have the potential to make the transport to the nucleus or cytosol more effective. This work had particularly focused on increasing the transduction efficiency by modifying particle's internally localized VP3 capsid protein with covalently bound membrane-penetrating peptides. Primary covalent genetic modification to the VP3 protein was the polyhistidine peptide KH27K. Its potential of improving the transduction effectivity was compared with two other peptide modifications - LAH4 and R8. The results of the transduction test showed that covalently bound R8 peptide had many-fold improved the transport to the nucleus when compared to the unmodified particles. The modification with LAH4 peptide had been regarded more effective only when was associated with the particles non-covalently.
